The core of the academic discourse surrounding advance directives (ADs) in dementia revolves around ethical considerations. Investigations into the lived experiences of individuals with dementia, particularly those affected by advertising, are surprisingly scarce, revealing a significant knowledge gap regarding the impact of national dementia-related legislation on these experiences. This paper considers the preparation phase of ADs in light of German dementia regulations. From 100 ADs and 25 episodic interviews with family members, we obtain the following results. Among 500 individuals recruited from a public assisted reproduction unit in Spain (502% female; 498% male; average age 361 years), FertiQoL was implemented. While a substantial amount of research has been dedicated to elucidating the diverse mechanisms of macroautophagy/autophagy in the last decade, a real-time assessment of this pathway is still a considerable challenge. The ATG4B protease, an early player in the activation cascade, prepares the autophagy key component MAP1LC3B/LC3B. Because live-cell reporting was inadequate for observing this phenomenon, we developed a FRET biosensor specifically designed to detect the priming of LC3B by ATG4B. A biosensor was crafted by incorporating LC3B flanked within a pH-resistant donor-acceptor FRET pair, Aquamarine-tdLanYFP. Our research demonstrates that this biosensor exhibits a dual-output capability. ATG4B's priming of LC3B, as indicated by FRET, is visually characterized by the spatial variations in priming activity, as observed through FRET imaging resolution. Determining the degree of autophagy activation is contingent upon quantifying the number of Aquamarine-LC3B puncta, secondarily. A decrease in ATG4B led to the accumulation of unprimed LC3B, and priming of the biosensor was not observed in ATG4B knockout cells. The priming deficiency can be ameliorated by the wild-type ATG4B or the partially active W142A mutant, but not by the catalytically inactive C74S mutant.
